I thought I might clarify a little..

The image mosaic plugin lets you use a mosaic as a data source. What data 
source you use inside of Geoserver doesn't really matter to GeoWebCache - all 
GeoWebCache sees is what Geoserver outputs as standard WMS responses, i.e. 
getcapabilities, getmap responses.

To clarify a bit further, not all WMS responses can be cached with GeoWebCache. 
It does cache tiles (i.e. getMap responses), but it doesn't cache other WMS 
responses such as GetFeatureInfo, GetLegendGraphic, etc; it proxies back to 
Geoserver for them.
