Hi folks,
THE PROBLEM
I am planning to express a growth factor as an Fc-fragment fusion in 293T cells
which I would transfect with the appropriate construct. My concern is using the
medium for the growth of my target cells. Filtration of the 293-conditioned
medium should be enough, but I am not 100% sure this would prevent occasional
events of contamination, and since the target cells would grow very slowly,
while 293T cells are fast, I wouldn't want to end up doing STR analysis for
whchever clone I get to grow.
THE QUESTION
I read somewhere of a 293T cells derivative which lacks part of a metabolic
pathway and would only grow if the medium is supplemented with some nutrients
that are not normally in media's formulae, but I cannot recall what their name
is. I googled "293T derivative" AND "conditional growth" and stuff like that,
but I am having a hard time browsing through non-relevant results. Anybody can
help?
Cheers
Alfredo
