Does this happen with just one file or does it happen with all files.
Lack of a PDF output option could mean that there its a problem with 
your LaTeX installation (or that LyX thinks there is).

As to the inability to save changes, could it be that the account 
under which you are running LyX lacks write permission in the 
directory where the configuration file lives? There also have 
been problems on Windows systems when the user its using
a roaming profile (home directory on a network server). I don't 
know if those issues have been cured in recent releases.

All else failing, you might try hiding your LyX user directory 
(by renaming it). The next time you start LyX, it will create a 
new one. I've heard of instances of a user profile becoming corrupted.
