The neat way would be to restore the bytes of the Live ISO to their
original state. In the simplest case that would be copying the ISO
image freshly to the USB stick.

A less invasive fix would be to find out where the changes sit in the
range of the ISO and to only restore those changed bytes. First suspect
would be the partition table block(s).
Which blocks are part of the table depends on the table type. With a
MBR/DOS partition table and ionly primary partitions it would only be
the first block of the USB stick. But with GPT it would be more blocks
near the start.

But maybe it wasn't only your partition table manipulations which
changed bytes in the ISO, but also boot firmware or other operating
systems which felt entitled to put files into the EFI System Partition
of the USB stick. See for example:
  https://bugs.debian.org/cgi-bin/bugreport.cgi?bug=1056998
In this case your USB stick ISO will change checksums again as soon as
you allow the manipulating systems access to it.

Have a nice day :)
