I dont know how to check this in windows. This could only be about the file on the server; it's samba trying to open the file, and samba isn't going to try to open files on the client. What are the permissions on the /var/lib/samba/printers/W32X86 directory? Note that the existing file on your filesystem is W32X86/3/SETUP32.DLL, but the error mesage is from trying to open W32X86/SETUP32.DLL.
