First, after a long time I started with KDE from Testing and after a
start of Icedove the first new mail is presented with a sound reminder.

Check your settings in your prefs.js if you have sound support enabled.
Also check your setting in "General" if the selected 'Use the following
sound file' is working, if not you have missconfigured soundsystem.
More informations you will find in [1].

If yes, please start with disabled plugins and in safe-mode, maybe there
is something that block the sound output.
If that doesn't work backup your current profile and start from scratch
with a fresh clean profile. Very offen problems comes in context with a
"old" profile.js migrated from the beginning of using Icedove. To be
sure the problem is a "real" software problem please check if your issue
is not depending on the old profile.

No, that's not true. We do no special handling here, the source isn't
modified here from the Debian side.

For me that isn't importend. I have static times there I check my mails,
I made the decision if I look at the mail, not the MTA.
My life is easier with that rule.

And, Icedove will only play a sound if the mail is the really first one
new in the queue and you have no others unread new messages.
If you have other ones that are unread and a new mail is comming no sound
will played.
