Got the bug today with a fresh install Karmic (20090916) Since in Karmic there are no xorg.conf file exists in the /etc/X11 directory anymore, nvidia-settins doesn't like that. :( (Tried create an empty xorg.conf file, no good)
In the end, you will need to run: sudo nvidia-xconfig Then, gksudo nvidia-settings Everything will work after that.
