The lock screen runs as a different user so it will use the system default settings rather than your own. You should be able to configure your preferences to make them system wide, in
Settings > Language & Region > Manage Installed Languages > Regional Formats > Apply System-Wide. As for: b'org.gnome.desktop.interface' b'clock-format' b"'12h'" that's probably a separate setting.
