A reboot won't be necessary, it should work after you log out and re-log
in. For some reason it is not grabbing the key when a grab is already
pending.

Is it possible that some app use same key-binding shortcuts? What is the
primary key here? Super? Try some other modifier like CTRL+SHIFT. Have
you used gnome-tweak-tool or compiz-config-settings-manager to add
custom shortcuts?

Do this on fresh install:

1) Open terminal on unity.
2) Run "unity-settings-daemon -r --debug", keep it open
3) Now Try removing the shortcut from Unity control center -> keyboard -> 
shortcuts
4) Try re-adding the shortcut.
5) Paste the output from terminal on https://pastebin.ubuntu.com/
