I'm not sure why this is marked invalid, but multiple Python installations will definitely break the printer configuration utility. I had the system supplied Python 2.6.5 in /usr/bin/python and Python 2.7 in /usr/local/bin/python and system-config-printer failed at startup when trying to import gobject until I removed Python 2.7 from my path.
If this restriction is documented somewhere, I haven't seen it.
