Does that mean the S loses its setting if you don't use it for 48
hours? Or only if the main batteries are removed for 48 hours?

I suspect that's confusion on the part of the websites. The D comes

a CR-2016, the DS does not, according to their respective manuals. The
DS uses a capacitor to power the settings storage, which lasts ~48
hours
while the D uses the lithium battery.
