If the hardware and the driver support it, then it should work. It's pretty easy to hack up /usr/xenocara/xserver/hw/vfb/InitOutput.c to support depths other than the stock 8, 15, 16, 24, and 30, (yes, 30, not 32), which is useful for testing if you don't have actual hardware with these modes.
