Hi, I am having some trouble with miRNA expression. I have not worked with miRNAs before. Can someone tell me how the 5p and 3p arms work with respect to a specific cell line? I know we can either have a mature 3p miRNA or a 5p miRNA. But how does it work when I over express miRNA using a plasmid? Can the over expression plasmid be targeted towards a specific mature sequence (3p or 5p)?
