hello everyone, I wanted to isolate plasma membrane from the neuronal cells using PERCOLL (sigma,osmolality max.25 mOsm/kg H2O, density 1.130+/- 0.005).
I need 30% final working percoll solution to isolate plasma membrane.So, in the product information sheet about percol said like we should mix percoll and required media in 9:1 ration and then use the below said given a formula, to get the final working percoll solution. Volume Calculation V0 = V[( a -0.1 )( b-0.9)] / (c-1) V0 = Volume of undiluted Percoll/Percoll PLUS required in ml V = Volume of final working solution in ml a = Desired density of final working solution c = Density of Percoll/Percoll PLUS undiluted b = Density of 1.5 M NaCl (1.058 g/ml) or 2.5 M sucrose (1.316 g/ml) So, can anybody, please help me to solve my problem like from 9:1 percol mix, how much volume should I take to get 30% final working percoll solution and what will be the density of my 30% percoll working solution.
