Go to Settings > Touchscreen > Touchscreen. Observe that it shows a device
named “Wacom HID 488F Finger” and the Enable checkbox works.

Go to Settings > Keyboard > Shortcuts. Observe that a section called “Wacom
Tablet” exists, it contains a shortcut named “Enable/Disable the Touch Tool”
and the shortcut set for it is the same as was set in KDE 5, at which time it
worked. Press the shortcut, nothing happens. Try normal things such as changing
the shortcut to something else, deleting the Wacom Tablet section etc., doesn’t
help.

Given that the Enable checkbox works fine, hopefully it’s not a complicated
hardware problem but just a question of the shortcut triggering the correct
action.
