The original poster said OpenPGP, not GnuPG, so -- I think PGP Corporation's PGP Desktop has to be considered. They use a mail proxy to prove transparent encryption and signing, and I've confirmed that it works with Entourage. (I haven't confirmed Outlook.) I personally don't like PGP Desktop for Mac, despite owning a license for it. I prefer having more fine-grained control over what messages I sign and encrypt. But if you absolutely must use it with Outlook or Entourage, PGP Desktop is probably the best bet. That said, PGP Desktop is not part of GPGTools, and is unlikely to ever be. :)
