Problem: I just discovered that thunderbird 52.4.0 (64bit) on Ubuntu Xenial can't play a wav file for new mail. The default new mail sound can be play though.
Solution: I installed "Sound Converter" from Ubuntu Software and used it to covert my .wav file to .ogg format (mentioned by @doxland) and tried it on Thunderbird. It works... :) Thanks @doxland for sharing about .ogg format. I did not know such a sound format had existed.
