By default, exim uses colon as its list separator. So "::1" is "empty item, :1", "0000:0000::1" is "0000, 0000, empty item, :1", and so on. Are you configuring the interface list via debconf, or by directly editing the configuration? afaics the debconf setup intentionally switches the list separator to be semi-colon in order to avoid such issues.
