http://packages.debian.org/search?searchon=contents&keywords=.supp&mode=path&suite=stable&arch=any reveals that of the various possible locations, /usr/lib/valgrind is favourite and /usr/share/doc is not used. I would put it in /usr/lib/valgrind, as this increases the motivation for default.supp to be automatically generated by an apt hook. It is also the only place I've ever thought of looking for .supp files myself.
