Yes with the # in front.
Probable it is /dev/sda1 and you only need to change that to
LABEL==dns1-rootfs
It is/was indeed:
# kopt=root=/dev/sda1 ro
Is it grub's feature or are they "helper comments" used by Debian tools?
Either way, I don't think it should touch old/existing entries; there
can be multiple different "root=" definitions in menu.lst file.
