For clarity, the most man page should explicitly indicate that the
user-level configuration file should be located at ~/.mostrc as I
surmised from /usr/share/doc/most/README. 

The manpage also refers to a "system configuration file", but does not
indicate where such a file should be placed nor does the most package
contain one. The README file is silent on this issue.
