Yes, it does look like it's missing a line such as:
 xxx:Debian testing/unstable (x86_64):/usr/bin/md5sum:/bin:

Here's what I got from os.dat:

 $ sudo grep "Debian" /var/lib/rkhunter/db/os.dat
 102:Debian testing/unstable (i386):/usr/bin/md5sum:/bin:
 103:Debian 2.2 (i386):/usr/bin/md5sum:/bin:
 105:Debian 3.0 (i386):/usr/bin/md5sum:/bin:
 142:Debian testing/unstable (sparc64):/usr/bin/md5sum:/bin:
 156:Debian 3.1 (i386):/usr/bin/md5sum:/bin:
 179:Debian 3.1 (x86_64):/usr/bin/md5sum:/bin:
