there are two init scripts to configure serial ports:
  /etc/init.d/setserial
  /etc/init.d/etc-setserial

In case of an existing /etc/serial.conf, the script /etc/init.d/setserial
can be only used to handle modules or get some status. The actual configuration will be done by /etc/init.d/etc-setserial. So your
  /etc/init.d/setserial start
should have been
  /etc/init.d/etc-setserial start

So if you don't mind I would close that bug and keep everything as it is
now.
