The Make manual says in the node "Guile Function":
You can determine whether GNU Guile support is available by checking
the '.FEATURES' variable for the word GUILE.
So this should do the trick:
$ make --eval="all:;@echo ${.FEATURES}"
Here it shows this:
target-specific order-only second-expansion else-if shortest-stem undefine
oneshell nocomment grouped-target extra-prereqs notintermediate shell-export
archives jobserver output-sync guile load dospaths
As you see, "guile" is right there.
